A new way of exploring immunity: Linking highly multiplexed antigen recognition to immune repertoire and phenotype

Single cell–based methods are the only way to directly observe antigen binding to T cells, while simultaneously sequencing the paired and recombined alpha and beta TCR genes. This is achieved by generating Single Cell 5' libraries and V(D)J enriched libraries, which provide TCR sequences, in combination with Feature Barcode technology, which uses highly multiplexed pMHC multimer reagents, each carrying a pMHC and a distinct molecular barcode, to identify binding specificities.
