Obtain your sample
Start with your frozen tissue of interest. Compatible sample types include mouse heart, liver, brain, and a variety of human tumors.

Break apart your frozen sample
Use the provided pestle and lysis buffer to mechanically disrupt the frozen tissue into a homogeneous mix.

Isolate nuclei
Pass the homogenized tissue through the provided spin column to isolate nuclei from the disrupted sample.

Remove debris
Clean up the nuclei suspension using the supplied debris removal solution and buffer washes.

Resuspend nuclei and QC
Resuspend the nuclei in the recommended resuspension buffer, check nuclei quality, and proceed to your single cell assay of choice.

Frequently asked questions
The Chromium Nuclei Isolation Kit has been validated with the following frozen tissues: mouse heart, liver, brain, lung, kidney, small intestine, and a variety of human tumors. An up-to-date list of tissues tested is available on the 10x Genomics Support site. This kit is not compatible with cell suspensions, plant or insect material, calcified tissues, or FFPE tissues.
Supported tissue sizes range from 3-50 mg per column. For pieces of tissue which are larger, break up the tissue into 50 mg pieces, run multiple dissociation columns, and combine nuclei at the final step. It may be possible to run pieces <3 mg, but yield is dependent on tissue type.
All the consumables and reagents needed to generate a clean nuclei suspension are included in the kit and utilize standard lab equipment. No specialized instrument is needed.
Compatible single cell assays include Chromium Single Cell ATAC, Single Cell Multiome ATAC + Gene Expression, Single Cell Gene Expression, and Single Cell Gene Expression Flex. Follow the nuclei resuspension recommendations for the specific assay you are running. No modifications to the single cell workflows are necessary.
10x Genomics provides analysis and visualization software to help you analyze your single cell data. Follow analysis recommendations for the single cell assay of interest. For single cell gene expression assays, make sure introns are included during analysis. You can download Cell Ranger and Loupe Browser software from the 10x Genomics Support site at no cost.
